RNA tumor viruses and human lymphomas.

چکیده
We have used molecular hybridization to detect virus-specific RNA in tumors ( I ) , and have found that corresponding neoplasias of murine and human origin exhibit remarkable similarities. Thus, human breast carcinomas contained (2) RNA possessing sequence homology to that of mouse mammary tumor virus (MMTV). This type of RNA was unique to the malignant adenoand medullary-carcinomas, being undetectable in normal breast tissue and in such benign pathologies as fibrocystic disease and fibroadenoma. In keeping with the known unrelatedness of the murine leukemogenic and mammary tumor viruses, we found that breast Cancer RNA did not hybridize to DNA complementary to the RNA of Rauscher leukemia virus (RLV). Finaly, and more compelling, was the demonstration that human leukemic cells (3) and human lymphomas (4), including Burkitt's disease (5), contain RNA showing homology to that of Rauscher leukemia virus and not to that of mouse mammary tumor virus.

an RNA related in sequence to that of the Rauscher leu kemia virus. The viral-related RNA found in these lym phomas is a 70 S component encapsulated with RNA-instructed DNA polymerase in a particle possessing a den sity characteristic of the RNA tumor viruses. Further more, the DNA synthesized by the lymphoma particles hy bridizes specifically to the RNA of the Rauscher leukemia virus. The part...
متن کاملHuman Reoviruses Serotype 3 Effectively Target Huh-7 Cells
Abstract: Background and Aims: Huh-7 is a cell line that was derived from a liver tumor of a Japanese man. Hepatocellular carcinoma (HCC) is considered as a primary liver cancer. Highly resistant tumor to treatment which causes the death of many patients annually. Thus, targeting the cancer cells by using a new method could be effective in...
متن کاملMolecular Approach to the Etiology of Human Cancer
DNA—RNA hybridization was used to explore whether human neoplasias contain RNA molecules having sequence homologies to those of the RNA tumor viruses known to cause similar diseases in animals. The pattern of specific RNA's found in the human tumors showed a remarkable concordance with the predictions deducible from the animal systems. Thus, human breast cancer contains RNA homologous only to t...
متن کاملRetroviruses as etiologic agents of some animal and human leukemias and lymphomas and as tools for elucidating the molecular mechanism of leukemogenesis.
I T HAS LONG BEEN KNOWN that retroviruses (RNA tumor viruses, type-C viruses, oncornaviruses, leukemia viruses) can induce a variety of neoplasias (leukemias and lymphomas, sarcomas, and occasionally carcinomas) in animals, and they have been identified as the prime factor in the pathogenesis of many naturally occurring leukemias and lymphomas in several animal species. Although there are recen...
متن کاملIn situ demonstration of Epstein-Barr virus small RNAs (EBER 1) in acquired immunodeficiency syndrome-related lymphomas: correlation with tumor morphology and primary site.
Some acquired immunodeficiency syndrome (AIDS)-related lymphomas (ARLs) are infected with Epstein-Barr virus (EBV), although the frequency and importance of this association is disputed. Using paraffin section RNA in situ hybridization (ISH) with digoxigenin-labeled riboprobes, we screened 16 central nervous system (CNS) non-Hodgkin's lymphomas (NHLs), 101 systemic NHLs, and 11 Hodgkin's diseas...
